Background of TYRO3 antibody
The transmembrane signalling by receptor tyrosine kinases invloves the activation of cell surface receptors as a result of ligand binding and subsequenct actviation of cytoplasmic tyrosine kinase domain that resides in the cytoplasmic loop. The tryrosine kinase receptors are critical for cell survival, proliferation and cell differentiation. The Tyro 3 family receptor members include Tyro 3 (also known as Sky, Brt, Tif, Rse, Dtk or Etk); Axl ½ (also known as Ark, Ufo, Tyro 7) and Mer (also known as Eyk, Nyk, tyro 12) (1-2). These receptors share distinctive structural domains in the extracellular region, composed of 2 immunoglobulin-related domains linked to 2 fibronectin type III repeats. These structural motifs are characteristics of cell adhesion molecules such as neuronal cell adhesion molecule (involved in proper migration of neuronal crest cells), and protein phosphatase-kappa and m.
The growth-arrest specific gene 6 (Gas 6), initially cloned form serum starved 3T3 fibroblast, causes cell cycle re-entry of serum starved 3T3 cells and also prevent them from apoptotic cell death caused by complete serum removal. The Gas 6 has been identified as a common ligand for all Tyro3 family receptor kinases including Mertk, tyro3 and Axl1/2. Triple mutant mouse (Tyro3-/-; Axl-/- ; Mer-/-) exhibit multiple major organ defects suggesting their involvement in differentiation and growth. Tyro3 receptors are widely expressed in neuronal tissue, lymphoid, vascular, reproductive and retinal tissues (3). The role of Tyro3 receptors in spermatogenesis is just emerging (4). Tyro3 is a 880-890 aa protein with multiple functional domains characteristics of receptor tyrosine kinases.
