Background of PRKCI antibody
This gene encodes a member ofThe protein kise C (PKC) family of serine/threonine protein kises.The PKC family comprises at least eight members, which are differentially expressed and are involved in a wide variety of cellular processes.This protein kise is calcium-independent and phospholipid-dependent. It is not activated by phorbolesters or diacylglycerol.This kise can be recruited to vesicle tubular clusters (VTCs) by direct interaction withThe small GTPase RAB2, whereThis kise phosphorylates glyceraldehyde-3-phosphate dehydrogese (GAPD/GAPDH) and plays a role in microtubule dymics inThe early secretory pathway.This kise is found to be necessary for BCL-ABL-mediated resistance to drug-induced apoptosis andTherefore protects leukemia cells against drug-induced apoptosis.There is a single exon pseudogene mapped on chromosome X. Western blot analysis of PKC iota/zeta (pT412/410) expression in MCF7 TNFa-treated (A); NIH3T3 LPS-treated (B); PC12 UV-treated (C) whole cell lysates.
Immunohistochemical analysis of PKC iota/zeta (pT412/410) staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugad compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Immunofluorescent analysis of PKC iota/zeta (pT412/410) staining in MCF7 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 deg C in a hidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
