Background of Nestin antibody
Nestin is a large intermediate filament protein originally identified by Hockfield and McKay (1985). It is predominately expressed in stem cells of the developing nervous system. Terminal differentiation is associated with a loss of nestin expression. Nestin expression has also been noted in other embryonic tissues, also in most Glioblastoma multiformes and many melanomas.
This cytoskeletal protein has been widely used as a predominant marker for stem/progenitor cells, glioma cells, and tumor endothelial cells. Also, it is a superior angiogenic marker to evaluate neovascularity of endothelial cells in tumors. These highly specific antibodies to human nestin can aid in characterizing progenitor cells differentiating into distinct lineages, in enhancing the therapeutic potential of human neural stem/progenitor cells in the treatment of CNS diseases or injury, and in identifying neuroepithelial tumor cells.

Anti-Nestin Antibody - Western Blot. Western blot using Affinity Purified anti-Nestin antibody shows detection of a band ~220 kD corresponding to mouse Nestin (arrowhead). Approximately 30 g of MEF whole cell lysate was separated by SDS-PAGE using a 4-20% gradient gel. After transfer onto nitrocellulose, the membrane was blocked and then probed with the primary antibody diluted to 1:2000 overnight at 4°C. The membrane was then washed and reacted with a 1:10000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] MX ( for 45 min at room temperature. IRDye800 fluorescence image was captured using the Ddyssey Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.

ELISA: Nestin Antibody
Immunofluorescence: Nestin Antibody (4D11) - Mixed cultures of neonatal rat neurons and glia stained with anti-Nestin (red), chicken antibody to Vimentin and DNA (DAPI stain, blue). Astrocytes and neuronal stem cells stain strongly and specifically in a clearly filamentous fashion with the Nestin (4D11) antibody. The filamentous staining pattern is as expected as both Nestin and Vimentin are components of 10nm filaments.

Formalin-fixed and paraffin-embedded human hepatocarcinoma reacted with Nestin Antibody (S1409), which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated.

Day 3 newborn mouse brain lysates (Lane 1) or adult mouse brain lysates (Lane 2) were resolved by electrophoresis, transferred to nitrocellulose, and probed with purified monoclonal anti-mouse/rat nestin (clone Rat-401) antibody. Proteins were visualized using an anti-mouse-IgG secondary conjugated to HRP and chemiluminescence detection.
