AGE and Ageing
Long term incubation of proteins with glucose can, through Schiff base and Amadori rearrangement, lead to the formation of Advanced Glycation End Products (AGE). Precipitates generated through crosslinking are recognizable by a characteristic brown colour and auto-fluorescence.
Using antibodies AGE has been found in human lens, in chronic nephropathic kidney and amyloidosis, in ateriosclerotic artery walls and in brain tissue from patients suffering from Morbus Alzheimer. These results suggest a potential role of AGE in ageing and associated diseases.
Receptors for AGE (RAGE)
RAGE, the receptor for AGE, is a surface transmembrane protein, which is able to recognize different AGE products. In normal tissue the expression of RAGE is low, but at sites of inflammation, diabetic complications, amyloidosis or in tumor tissue expression is upregulated. It has been shown, that activation of RAGE may lead to cellular dysfunction by means of formation and precipitation of AGE products.
