Cullin-1, 2, and 3 are members of the family of human cullin genes (CUL-1, -2, -3, -4a, -4b, and -5) homologous to the S. cerevisiae cdc53 gene. Cullin-1 forms a complex with human p19skp1 and F box protein p45skp2, both of which, together with cdc53 (known as CUL A), as three subunits, form a ubiquitin ligase controlling proteolysis of G1 cell cycle regulatory proteins. The CUL-1/p19skp1/p45skp2 complex is thought to play a role in the ubiquitination of G1 regulatory proteins. Instead, Cullin-2 forms a complex with an inactive transcriptional elongation complex, SIII, formed by three subunits: Elongain C, Elongin B and VHL. The function of this association remains unknown. Cullin-3 is proposed to be a part of E3 proteinubiquitin ligase. It forms a tight complex with cyclin E and targets it for ubiquitination, which leads to its degradation by protesomes. Cyclin E is an activator of cdk2, which leads the cell to S phase.

Cul-3-/- mice show increased accumulation of Cyclin E protein and disruption of normal cell cycle regulation in vivo. Thus Cullin-3 is important in the regulation of S phase of cell cycle.

Cullins 4a, 5 and 7 are also essential components of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin ligase complex.
